ITX-0121 is a small molecule drug candidate developed as a targeted therapy for cancers deficient in transcription-coupled nucleotide excision repair (TC-NER). The drug acts by blocking the TC-NER polymerase complex, leading to its disengagement from DNA. This disruption initiates an irreversible process of apoptosis (cellular suicide) in cancer cells that are unable to properly repair DNA damage due to deficiencies in their DNA damage response (DDR) mechanisms. As many cancers exhibit such deficiencies, ITX-0121 represents a potential therapeutic strategy specifically for tumors with impaired TC-NER pathways[1][2][4].
