ITYR-DBRMD is a first-in-class small-molecule inhibitor of iodothyronine deiodinase type 3 (DIO3), developed by Dio Tree Ltd for the treatment of cancer, specifically platinum-resistant ovarian cancer. DIO3 is an enzyme that inactivates triiodothyronine (T3), a thyroid hormone that normally exerts tumor-suppressive effects by inhibiting proliferative and DNA-damage stress pathways. By selectively inhibiting DIO3, ITYR-DBRMD restores intracellular T3 levels and enhances tumor suppression. Preclinical studies have demonstrated that ITYR-DBRMD exhibits robust antitumor activity as a monotherapy and synergistic effects when combined with carboplatin, while maintaining a favorable safety profile and utilizing albumin-based delivery for enhanced tumor retention.
