IUB288 is a long-acting, water-soluble, and highly selective glucagon receptor agonist developed for research into metabolic diseases such as obesity and diabetes. It acts by stimulating the glucagon receptor (GCGR), leading to increased energy expenditure, reduced fat mass, and weight loss in preclinical models. Mechanistically, IUB288 increases hepatic fibroblast growth factor 21 (FGF21) expression and plasma levels, enhances fatty acid oxidation via farnesoid X receptor (FXR) signaling in the liver, and augments insulin action through increased hepatic AKT phosphorylation. Chronic administration of IUB288 results in significant reductions in body weight primarily due to fat loss rather than lean mass catabolism. The drug has been used extensively in animal studies to elucidate the role of GCGR signaling in energy metabolism and its potential as an anti-obesity therapy[2][3][5][6].
