Drug intelligence / Profile preview
IV3 is a preclinical small molecule inhibitor of the YAP1/TAZ-TEAD interaction, originally developed by Inventiva. It acts by binding to the TEAD Ω-loop pocket, thereby preventing the formation of the oncogenic YAP1/TAZ-TEAD transcriptional complex and inhibiting the expression of downstream target genes such as CTGF and CYR61. In preclinical studies, IV3 has demonstrated the ability to inhibit cell proliferation in meningioma and other Hippo pathway-driven cancer models, both as a monotherapy and in combination with MAPKinase or Pi3K-AKT-mTOR inhibitors. In early 2025, Inventiva announced a pipeline prioritization plan to focus exclusively on its lead MASH candidate, lanifibranor, resulting in the termination of its preclinical YAP-TEAD program, including the development of IV3.
