This combination therapy consists of ivarmacitinib, a selective Janus kinase 1 (JAK1) inhibitor; apatinib, a small-molecule tyrosine kinase inhibitor targeting vascular endothelial growth factor receptor 2 (VEGFR2); and camrelizumab, a humanized monoclonal antibody against programmed cell death protein 1 (PD-1). Developed for the treatment of unresectable advanced hepatocellular carcinoma (uHCC), particularly in patients who have developed acquired resistance to first-line immune checkpoint inhibitors, this triple regimen aims to reshape the tumor immune microenvironment. By inhibiting JAK1, the therapy seeks to reduce immunosuppressive cell infiltration (like MDSCs) and restore T-cell function, while apatinib provides anti-angiogenic effects and camrelizumab reactivates the immune response against tumor cells. The regimen is currently being evaluated in a Phase 2 clinical trial sponsored by the First Affiliated Hospital of Chongqing Medical University.
