Ivicentamab (GEN3009) is a humanized IgG1 bispecific monoclonal antibody that targets two distinct epitopes on the CD37 antigen. It incorporates an E430G hexamerization-enhancing mutation to promote the formation of antibody hexamers at the surface of CD37-expressing tumor cells. This design enhances complement-dependent cytotoxicity (CDC) and also mediates antibody-dependent cell-mediated cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). The drug was developed using Genmab’s DuoBody and HexaBody technologies. Its primary indication was for relapsed or refractory B-cell non-Hodgkin lymphoma, including subtypes such as diffuse large B-cell lymphoma (DLBCL), follicular lymphoma (FL), chronic lymphocytic leukemia (CLL), and small lymphocytic lymphoma (SLL)[1][2][6].
