IVIV 378 is a monovalent polysaccharide-protein conjugate vaccine candidate developed by InventVacc Biologicals in collaboration with the National Research Council of Canada (NRC) and the Public Health Agency of Canada (PHAC). It is designed to prevent invasive *Haemophilus influenzae* type a (Hia) infections, including meningitis, sepsis, and pneumonia, which have emerged as significant health threats in Indigenous communities in northern Canada and Alaska. The vaccine consists of the Hia capsular polysaccharide (CPS) chemically conjugated to a recombinant CRM197 (rCRM197) carrier protein using a hydrazide-polyethylene glycol-hydrazide linker. This conjugation strategy is intended to induce a T-cell dependent immune response, providing long-lasting protection and immunologic memory. Formulated with the aluminum phosphate adjuvant AdjuPhos, IVIV 378 is currently undergoing Phase 1 clinical evaluation to assess its safety and immunogenicity in healthy adults.
