Overview

Ivocilimab (AK131) is a humanized bifunctional fusion protein, also classified as an immunocytokine, developed by Akeso Biopharma. It is engineered to simultaneously target the programmed cell death protein 1 (PD-1) and the interleukin-15 (IL-15) signaling pathway. The molecule consists of a monoclonal antibody against PD-1 fused to an IL-15 variant (typically an IL-15/IL-15Rα complex). This dual-action mechanism is designed to block the immunosuppressive PD-1/PD-L1 checkpoint while providing localized activation of IL-15 receptors on T cells and natural killer (NK) cells within the tumor microenvironment. By co-delivering IL-15 to PD-1-expressing immune cells, ivocilimab aims to enhance their proliferation, survival, and effector functions, potentially overcoming resistance to conventional PD-1/PD-L1 inhibitors. It is currently under clinical investigation for the treatment of advanced solid tumors, including non-small cell lung cancer (NSCLC).
