Overview

This drug combination consists of two bispecific antibodies, ivonescimab and cadonilimab, administered alongside etoposide and carboplatin (EC) chemotherapy. Ivonescimab (AK112) is a first-in-class bispecific antibody that simultaneously targets programmed cell death protein 1 (PD-1) and vascular endothelial growth factor (VEGF). Cadonilimab (AK104) is a bispecific antibody targeting PD-1 and cytotoxic T-lymphocyte-associated protein 4 (CTLA-4). This combination is being investigated in a Phase II clinical trial sponsored by Shanghai Pulmonary Hospital for the treatment of extensive-stage small cell lung cancer (ES-SCLC). The regimen aims to leverage dual checkpoint inhibition and anti-angiogenesis to enhance anti-tumor activity, while the chemotherapy components (etoposide and carboplatin) provide cytotoxic effects through topoisomerase II inhibition and DNA alkylation.
