Drug intelligence / Profile preview
Ivonescimab plus leucovorin, fluorouracil, and oxaliplatin is an investigational combination regimen consisting of the novel bispecific antibody ivonescimab and the standard mFOLFOX6 chemotherapy regimen. Ivonescimab is a first-in-class tetrameric bispecific antibody that simultaneously targets programmed cell death protein 1 (PD-1) and vascular endothelial growth factor (VEGF), combining immunotherapy with anti-angiogenesis. The chemotherapy backbone consists of oxaliplatin (a platinum-based DNA-crosslinking agent), fluorouracil (a thymidylate synthase inhibitor), and leucovorin (calcium folinate, a folate analog that enhances fluorouracil's efficacy). This combination is primarily being developed by Summit Therapeutics and Akeso for the first-line treatment of patients with unresectable or metastatic colorectal cancer (mCRC), and is currently being evaluated in the global Phase III HARMONi-GI3 trial.
