Ivonescimab + olaparib is an investigational combination therapy consisting of ivonescimab, a bispecific monoclonal antibody targeting programmed death-1 (PD-1) and vascular endothelial growth factor A (VEGF-A), and olaparib, a small molecule inhibitor of poly ADP ribose polymerase (PARP). Ivonescimab enhances anti-tumor immune responses by blocking the PD-1 checkpoint and impairs tumor angiogenesis by inhibiting VEGF-A. Olaparib induces cancer cell death by inhibiting DNA repair in cancer cells reliant on PARP function, particularly those with BRCA1/2 mutations. This combination aims to synergize immune activation, anti-angiogenic effects, and synthetic lethality in tumors, especially non-small cell lung cancer (NSCLC), by combining modalities of immunotherapy and DNA damage-based targeted therapy[2][4][6].
