Overview
This is a **combination investigational therapy** comprising **ivonescimab** (a bispecific monoclonal antibody targeting PD-1 and VEGF) and **TAS-102** (an oral combination of trifluridine and tipiracil, marketed as Lonsurf). Ivonescimab acts by blocking both the PD-1 immune checkpoint and VEGF-mediated pathways, aiming to enhance immune response and inhibit angiogenesis in tumors. TAS-102 includes trifluridine, a nucleoside analog that incorporates into DNA and disrupts DNA function, and tipiracil, which inhibits thymidine phosphorylase to maintain trifluridine exposure. This combination is being studied for the treatment of patients with advanced or metastatic solid tumors, including colorectal cancer, especially in cases refractory to standard therapies. The combination is in clinical trials, including a phase II study investigating its efficacy and safety[6][7][10]. Ivonescimab is under development by Summit Therapeutics and Akeso, while TAS-102 (Lonsurf) is developed by Taiho Pharmaceutical and marketed in some regions by Servier[5].
