Overview

Ivosidenib is a small molecule inhibitor that selectively targets the mutant isocitrate dehydrogenase 1 (IDH1) enzyme. By inhibiting mutant IDH1, it reduces the production of the oncometabolite D-2-hydroxyglutarate, which can promote tumorigenesis. Ivosidenib is primarily indicated for the treatment of adult patients with relapsed or refractory acute myeloid leukemia (AML) harboring an IDH1 mutation and has also been approved for use in patients with previously treated, locally advanced or metastatic cholangiocarcinoma with an IDH1 mutation[6][7]. The drug was developed by Agios Pharmaceuticals and commercialized by Servier.
