Overview
IVR-103 is a sustained-release (SR) formulation of avacincaptad pegol, a pegylated RNA aptamer that functions as a complement C5 inhibitor. Developed by Iveric Bio (now a subsidiary of Astellas Pharma), it is designed for the treatment of geographic atrophy (GA) secondary to age-related macular degeneration (AMD). The formulation utilizes a sustained-release delivery system, such as a biodegradable silica-based matrix, to provide prolonged therapeutic levels of the drug in the vitreous. This approach is intended to reduce the frequency of intravitreal injections compared to the immediate-release formulation, Izervay, thereby improving patient compliance and reducing the treatment burden. IVR-103 works by binding to complement protein C5 and preventing its cleavage into C5a and C5b, which inhibits the formation of the membrane attack complex (MAC) and slows the progression of retinal cell death.
