Overview

Ixabepilone + vorinostat is an investigational combination therapy evaluated primarily for metastatic breast cancer. Ixabepilone is a semisynthetic analog of epothilone B and acts as a microtubule inhibitor, disrupting cell division by stabilizing microtubules and preventing their depolymerization. Vorinostat (also known as suberoylanilide hydroxamic acid or SAHA) is a histone deacetylase (HDAC) inhibitor that induces acetylation of histones and non-histone proteins, leading to altered gene expression, cell cycle arrest, and apoptosis in cancer cells. Preclinical studies demonstrated synergistic antitumor activity when these agents are combined, likely due to enhanced disruption of cellular processes critical for tumor growth and survival. Clinical trials have established the maximum tolerated dose for this combination in patients with previously treated metastatic breast cancer, showing promising clinical activity with manageable toxicity profiles[1][3][8].
