ixazomib

Ixazomib is an oral small molecule proteasome inhibitor used primarily in the treatment of multiple myeloma. It works by selectively inhibiting the 20S proteasome, disrupting protein degradation within cancer cells, leading to apoptosis and inhibition of tumor growth. Ixazomib is notable as the first oral proteasome inhibitor approved for clinical use. It is typically administered in combination with lenalidomide and dexamethasone for patients with relapsed or refractory multiple myeloma. The drug has also been studied for other plasma cell disorders such as Waldenström's macroglobulinemia (off-label) and systemic light chain (AL) amyloidosis. Ixazomib was developed by Millennium Pharmaceuticals Inc and is manufactured by Takeda Pharmaceutical Company. It received FDA approval in 2015 and EMA approval in 2016; it was approved in China in 2018.
