Overview

Ixazomib + thalidomide + dexamethasone is an all-oral combination regimen used primarily for the treatment of relapsed or refractory multiple myeloma. Ixazomib is a small molecule proteasome inhibitor that disrupts protein degradation in cancer cells, leading to apoptosis. Thalidomide acts as an immunomodulatory agent, binding to cereblon and promoting the degradation of transcription factors IKZF1 and IKZF3, which are essential for myeloma cell survival. Dexamethasone is a synthetic glucocorticoid with anti-inflammatory and cytotoxic effects on lymphoid cells, enhancing the efficacy of the other agents. This combination has demonstrated clinical activity and tolerability in patients who have received prior therapies for multiple myeloma[1][3][4][8].
