Drug intelligence / Profile preview
Ixokallipin is a novel plasma kallikrein (KLK) inhibitor belonging to the serpin family, originally identified in the saliva of the deer tick *Ixodes scapularis*. It specifically targets and inhibits plasma kallikrein and coagulation factor XIIa (FXIIa), thereby blocking the intrinsic pathway of blood coagulation. Ixokallipin binds to endothelial cell and keratinocyte membranes, primarily via phosphatidic acid, and inhibits kallikrein-induced wound healing and keratinocyte migration. In vivo, it has been shown to inhibit venous thrombosis without causing significant bleeding, while also delaying skin wound healing by reducing cellular focal adhesions and membrane protrusions.
