ixotatug vedotin

**Ixotatug vedotin (TORL-1-23)** is a humanized monoclonal antibody-drug conjugate (ADC) targeting **claudin 6 (CLDN6)**, a tight junction protein overexpressed in certain solid tumors but absent in healthy adult tissues. It comprises an anti-CLDN6 antibody conjugated to **monomethyl auristatin E (MMAE)**, a microtubule-disrupting payload, via a protease-cleavable linker with a drug-antibody ratio (DAR) of 4. Developed by **TORL Biotherapeutics**, it is administered intravenously every 3 weeks and has shown promising antitumor activity in heavily pretreated **CLDN6-positive platinum-resistant ovarian cancer (PROC)** patients, with objective response rates up to 50% at higher doses (2.4-3.0 mg/kg), alongside a manageable safety profile primarily featuring grade 1-2 fatigue, peripheral neuropathy, and alopecia.[1][3][7]
