Proxovex-1 + pembrolizumab is a combination investigational therapy consisting of Ixovex-1, a novel oncolytic E1B-mutated recombinant adenovirus, and pembrolizumab, a monoclonal antibody checkpoint inhibitor targeting PD-1. Ixovex-1 selectively replicates within cancer cells by modulating E1B mRNA isoforms, thereby preferentially lysing tumor cells and stimulating local immune activation. Pembrolizumab blocks the PD-1 immune checkpoint, restoring T-cell activity against tumor cells. Early-phase clinical trials are evaluating this combination primarily for patients with advanced, unresectable, or metastatic solid tumors where standard therapies have failed[1][3]. Ixovex-1’s tumor-selective lysis may enhance the immunogenicity of the tumor microenvironment, potentially potentiating the antitumor activity of pembrolizumab.
