izalontamab

Izalontamab (SI-B001) is a tetravalent bispecific monoclonal antibody targeting both the epidermal growth factor receptor (EGFR) and human epidermal growth factor receptor 3 (HER3/ERBB3). Developed using SystImmune’s Specificity Enhanced Bi-specific Antibody (SEBA) platform, it simultaneously binds to EGFR and HER3 on cancer cells, blocking their phosphorylation and activation. This dual inhibition disrupts downstream signaling pathways critical for tumor cell proliferation and survival. In addition to direct signal blockade, SI-B001 leverages its wild-type Fc region to mediate innate immune effector functions against cancer cells. The drug is being developed primarily for the treatment of solid tumors with aberrant EGFR/HER3 expression, including non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), colorectal cancer, esophageal cancer, nasopharyngeal cancer, triple negative breast cancer, and other epithelial malignancies.
