Overview

Izalontamab brengitecan is a tetravalent bispecific antibody-drug conjugate (ADC) that targets both the Epidermal Growth Factor Receptor (EGFR) and Human Epidermal Growth Factor Receptor 3 (HER3). The drug is designed to bind to these receptors, which are frequently overexpressed in epithelial tumors, block their proliferative signaling, and deliver a cytotoxic topoisomerase I inhibitor payload to cancer cells. Upon receptor-mediated internalization, the conjugate induces genotoxic stress in the lysosome, resulting in apoptosis of tumor cells. It is being developed primarily for treatment of HER2-negative or HER2-low breast cancer, as well as small cell and non-small cell lung cancers, among other epithelial tumors. Izalontamab brengitecan is currently in Phase 3 and Phase 2 clinical trials for various advanced solid tumor indications.
