Overview

This is a **combination therapy** consisting of izalontamab brengitecan (not yet widely characterized in the literature), pembrolizumab, a PD-1 immune checkpoint inhibitor, and bevacizumab, an anti-VEGF monoclonal antibody. Pembrolizumab works by blocking the programmed cell death protein-1 (PD-1) receptor, releasing the brakes on anti-tumor cytotoxic T cells. Bevacizumab inhibits vascular endothelial growth factor A (VEGF-A), suppressing tumor angiogenesis and normalizing tumor vasculature, thereby potentially enhancing both delivery of anticancer drugs and immune cell infiltration to tumor sites. Izalontamab brengitecan, by its name, appears to be an antibody-drug conjugate (ADC), which consists of izalontamab (an antibody) linked to brengitecan (likely a cytotoxic payload related to camptothecin or SN-38, a topoisomerase I inhibitor), though precise details of its mechanism are not publicly available.
