Overview

Izastobart (TJ210001) is a fully human monoclonal antibody targeting the C5a receptor (C5aR, also known as CD88), developed by I-Mab Biopharma. It is designed to block the interaction between the anaphylatoxin C5a and its receptor on myeloid cells. In the tumor microenvironment, the C5a/C5aR axis is a key driver of immune suppression, as it recruits myeloid-derived suppressor cells (MDSCs), neutrophils, and M2 macrophages that inhibit T-cell activity. By antagonizing C5aR, izastobart aims to modulate the tumor microenvironment to be more pro-inflammatory and enhance the efficacy of other immunotherapies, such as PD-1/PD-L1 inhibitors. It is currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors.
