IMU-935 (izumerogant) is an orally available small molecule drug under development for the treatment of autoimmune and inflammatory diseases. It acts as a highly potent and selective inverse agonist of the retinoic acid receptor-related orphan nuclear receptor gamma truncated (RORγt), a key transcription factor driving Th17 cell differentiation and the production of pro-inflammatory cytokines such as IL-17A, IL-17F, and IFNγ. In addition to its primary action on RORγt, IMU-935 also inhibits dihydroorotate dehydrogenase (DHODH), providing a synergistic effect in reducing cytokine release. Preclinical studies have demonstrated strong inhibition of Th1 and Th17 responses without affecting thymocyte maturation. The drug has shown efficacy in animal models for psoriasis, colitis, multiple sclerosis, graft-versus-host disease, and inflammatory bowel disease. Clinical trials are ongoing for indications including psoriasis and castration-resistant prostate cancer[1][2][4][5][6][7].
