Overview

Izuralimab (formerly known as XmAb23104 or XmAb104) is a humanized Fc-engineered bispecific monoclonal antibody developed by Xencor. It targets both the programmed cell death 1 receptor (PD-1), an immune checkpoint inhibitor, and the inducible T-cell co-stimulator protein (ICOS/CD278), a co-stimulatory molecule on T cells. By simultaneously blocking PD-1 and activating ICOS, izuralimab aims to enhance anti-tumor immune responses through dual checkpoint inhibition and T-cell activation. The drug is designed with an engineered Fc domain to eliminate Fc gamma receptor binding in order to prevent unwanted depletion or activation of T cells by other immune cells[1][2][4]. Izuralimab has been investigated primarily for advanced solid tumors including melanoma and sarcoma but development in these indications has been discontinued[1][9].
