Overview

J147 is an experimental small molecule drug developed as a neuroprotective and neurotrophic agent. It is a derivative of curcumin and was initially designed to treat neurodegenerative conditions associated with aging, particularly Alzheimer's disease. Unlike most Alzheimer's drugs that target amyloid plaques, J147 was discovered by screening for anti-aging effects and targets age-related pathways. Its primary mechanism involves binding to mitochondrial ATP synthase (specifically the alpha-F1 subunit), modulating mitochondrial function and cellular energy metabolism. Preclinical studies in mice have shown that J147 can improve memory and cognition, reduce soluble amyloid-beta levels, increase brain-derived neurotrophic factor (BDNF), enhance synaptic markers, decrease inflammation markers (such as TNFα), and promote healthier blood vessels in the brain. It has also demonstrated benefits in models of diabetic neuropathy and increased lifespan in Drosophila flies[1][2][3][4][5][6].
