JA-003 is a selective monoclonal antibody designed to inhibit the transforming growth factor beta 1 (TGF-β1) and transforming growth factor beta 3 (TGF-β3) isoforms. Developed by Jace Therapeutics, the drug is engineered to provide potent inhibition of these specific isoforms while sparing TGF-β2, a strategy intended to mitigate the cardiotoxicity and other adverse effects historically associated with pan-TGF-β inhibition. TGF-β is a pleiotropic cytokine that plays a critical role in immune evasion, epithelial-to-mesenchymal transition (EMT), and fibrosis within the tumor microenvironment. By selectively blocking TGF-β1 and TGF-β3, JA-003 aims to restore anti-tumor immune responses and enhance the efficacy of other immunotherapies, such as checkpoint inhibitors, in patients with advanced solid tumors.
