Overview

JAA-F11-ADC (also known as hJAA-F11-DM1) is a preclinical antibody-drug conjugate (ADC) developed by For-Robin, a spinout of the University at Buffalo, for the treatment of solid tumors, specifically breast cancer (including triple-negative breast cancer) and lung cancer. The ADC consists of hJAA-F11, a humanized IgG1 monoclonal antibody that specifically targets the alpha-linked Thomsen-Friedenreich glycoantigen (TF-Ag-alpha), conjugated to the microtubule inhibitor payload DM1 (maytansine) via a maleimidocaproyl (MC) linker. TF-Ag-alpha is a tumor-associated carbohydrate antigen expressed on approximately 85% of human carcinomas but cryptic on normal tissues, where only the beta-linked form is present. Upon binding to TF-Ag-alpha on the cancer cell surface, JAA-F11-ADC is rapidly internalized, releasing DM1 to disrupt microtubule dynamics, arrest the cell cycle, and induce apoptosis.
