JAB-22000 is a potent, small molecule inhibitor specifically targeting the KRAS G12D mutation, developed by Jacobio Pharmaceuticals. KRAS G12D is a frequent oncogenic driver in several high-prevalence malignancies, including pancreatic ductal adenocarcinoma, colorectal cancer, and non-small cell lung cancer. JAB-22000 is designed to selectively inhibit the signaling activity of the mutant KRAS protein, thereby disrupting downstream pathways essential for tumor growth and survival. Beyond its potential as a standalone therapeutic, JAB-22000 is utilized as the cytotoxic payload in Jacobio's JAB-BX600, an antibody-drug conjugate (ADC) that targets EGFR to deliver the KRAS inhibitor directly to tumor cells. As of late 2023, the program was in the preclinical stage with IND filing anticipated for the 2023-2024 period.
