JAB-23E73 + nab-paclitaxel + gemcitabine is an investigational combination regimen being evaluated for the treatment of metastatic pancreatic ductal adenocarcinoma (PDAC) harboring KRAS gene alterations. JAB-23E73 is a proprietary, orally administered small molecule pan-KRAS inhibitor developed by Jacobio Pharmaceuticals. It is designed to target multiple KRAS mutation subtypes by inhibiting both the active ('ON') and inactive ('OFF') states of the protein with high selectivity, while sparing HRAS and NRAS. Nab-paclitaxel (Abraxane) is an albumin-bound nanoparticle formulation of paclitaxel that inhibits microtubule dynamics, and gemcitabine is a nucleoside analog that interferes with DNA synthesis. This combination therapy aims to provide synergistic antitumor activity by simultaneously targeting the primary oncogenic driver (KRAS) and utilizing standard-of-care cytotoxic chemotherapy in treatment-naive patients.
