JAB-24114 is a self-developed small molecule prodrug by Jacobio Pharma, designed as an inhibitor of glutamine-utilizing enzymes (GUEs). It acts by inhibiting multiple GUEs, thereby blocking several glutamine metabolism pathways that are essential for tumor growth, including glutaminolysis, nucleotide synthesis, hexosamine synthesis, coenzyme synthesis, and amino acid synthesis. Unlike selective glutaminase inhibitors that only block the conversion of glutamine to glutamate, JAB-24114 targets a broader range of metabolic processes. The drug is a prodrug of L-6-Diazo-5-oxo-norleucine (DON), which is preferentially distributed in tumors and activated there to minimize gastrointestinal toxicity associated with DON itself. Preclinical studies have shown strong antitumor effects and enhanced T cell function. JAB-24114 also demonstrates synergistic potential when combined with anti-PD-(L)1 antibodies or other targeted therapies such as SHP2 or KRAS inhibitors[1][3][4][5][8].
