JAB-26766 is a potent, orally bioavailable small molecule inhibitor of poly(ADP-ribose) polymerase 7 (PARP7), developed by Jacobio Pharmaceuticals. It demonstrates high selectivity for PARP7 over other PARP family members (>1800-fold selectivity over PARP2). Mechanistically, JAB-26766 inhibits the enzymatic activity of PARP7, leading to increased IFN-β secretion and phosphorylation of STAT1 in tumor cells. This results in enhanced anti-tumor immune responses and direct inhibition of tumor growth. Preclinical studies have shown significant antitumor activity across various solid tumor models in mice. The drug is being investigated primarily for advanced solid tumors and has potential for combination therapy with STING agonists or PD-(L)1 monoclonal antibodies[1][2][5].
