JAB-27670 is a highly potent, non-cyclic dinucleotide (non-CDN) small-molecule agonist of the stimulator of interferon genes (STING) pathway. Developed by Jacobio Pharmaceuticals, JAB-27670 is designed with sub-nanomolar activity and favorable water solubility, making it suitable for use as an immunostimulatory payload in antibody-drug conjugates (iADCs). It has been utilized in preclinical candidates such as JAB-X1800 (a CD73-targeted STING iADC) and JAB-BX400 (a HER2-targeted STING iADC). By targeting STING, JAB-27670 stimulates the production of type I interferons (such as IFN-beta) and pro-inflammatory cytokines, converting immunologically 'cold' tumors into 'hot' tumors to enhance anti-tumor immunity while minimizing systemic toxicity when delivered via targeted conjugates.
