JAB-30300 is an orally bioavailable small molecule developed by Jacobio Pharmaceuticals for the treatment of solid tumors harboring the P53 Y220C mutation. It acts as a P53 Y220C activator, binding with high affinity to mutant p53 proteins and restoring their tumor suppressor function. Preclinical studies have demonstrated that JAB-30300 induces tumor regression in multiple cancer models, including gastric, ovarian, breast, and lung cancers. The drug has also shown synergistic effects when combined with chemotherapy or oncogenic protein inhibitors. As of 2024, it is one of the first P53 Y220C activators to enter clinical trials globally[1][2][3][5][8].
