JAB-3068 is an orally bioavailable, small molecule inhibitor of protein tyrosine phosphatase non-receptor type 11 (PTPN11), also known as SHP2. It is designed to target, bind to, and inhibit the activity of SHP2, a key regulator in the Ras-Raf-MEK-ERK signaling pathway that controls cell survival, differentiation, and proliferation. By inhibiting SHP2-mediated signaling—including MAPK pathway activation—JAB-3068 aims to suppress tumor growth in cancers where this pathway is hyperactivated due to mutations or rearrangements. Additionally, it may modulate immune checkpoint pathways such as PD1-mediated signal transduction. The drug has been developed primarily for antineoplastic indications including non-small cell lung cancer (NSCLC), squamous cell carcinoma (including head and neck and esophageal subtypes), and other advanced solid tumors[1][3][4][7].
