**JAB-3312 + binimetinib** is an investigational combination therapy comprising JAB-3312, an allosteric inhibitor of SHP2 (Src homology region 2-containing protein tyrosine phosphatase 2), and binimetinib, a selective MEK inhibitor. JAB-3312 acts by inhibiting SHP2, a key mediator within the RTK/RAS/MAPK signaling pathway, which is often aberrantly activated in various cancers. Mechanistically, combining SHP2 inhibition with MEK inhibition (via binimetinib) aims to prevent pathway reactivation, overcome resistance encountered with single-agent targeted therapies, and enhance anti-tumor efficacy. The combination is being developed and tested for advanced solid tumors, particularly in patients with KRAS-mutant cancers[5][3].
