JAB-BX400 is a preclinical-stage investigational antibody-drug conjugate (ADC) developed by Jacobio Pharma. It is an immune-stimulating ADC (iADC) that targets the human epidermal growth factor receptor 2 (HER2) and delivers a stimulator of interferon genes (STING) agonist as its payload. The design aims to selectively deliver the STING agonist into HER2-expressing tumor cells, thereby locally activating innate and adaptive antitumor immune responses while minimizing systemic immune-related toxicity. Preclinical studies have shown that JAB-BX400 is effective against DS8201-resistant tumor models and in "cold" tumors such as the SK-OV-3 xenograft model, which are typically less responsive to immunotherapy[2][4][5][6][7]. The drug barely releases free payload in plasma, reducing off-target effects and cytokine release compared to competitors[7]. An IND application for clinical trials is planned for 2024–2025[2].
