JAB-BX467 is an immune-stimulating antibody-drug conjugate (iADC) developed by Jacobio Pharmaceuticals. It is designed to treat HER2-expressing solid tumors by combining a human epidermal growth factor receptor 2 (HER2)-targeting antibody with a small-molecule stimulator of interferon genes (STING) agonist payload. The drug's mechanism of action involves the targeted delivery of the STING agonist to the tumor microenvironment, where it activates the innate immune system, converting immunologically "cold" tumors into "hot" tumors. This approach is intended to address the high percentage of patients who do not respond to standard PD-1/PD-L1 checkpoint inhibitors. Preclinical studies have demonstrated that JAB-BX467 is stable, induces significantly less IL-6 compared to competitors, and can persistently eradicate tumor growth while establishing a strong immune memory effect. The candidate is currently in the IND-enabling stage, with an IND filing anticipated in the second half of 2026.
