JAB-X1800

JAB-X1800 is an immunostimulatory antibody-drug conjugate (iADC) developed by Jacobio Pharmaceuticals. It is composed of a monoclonal antibody targeting CD73 (5'-nucleotidase) conjugated to JAB-27670, a potent non-cyclic dinucleotide (non-CDN) STING (Stimulator of Interferon Genes) agonist payload. By targeting CD73, which is frequently overexpressed in the tumor microenvironment, JAB-X1800 selectively delivers the STING agonist to tumor cells and tumor-associated immune cells. This targeted activation of the STING pathway is intended to induce the production of Type I interferons and pro-inflammatory cytokines, thereby converting immunologically "cold" tumors into "hot" tumors and enhancing anti-tumor immune responses. Preclinical studies presented at the 2023 AACR Annual Meeting demonstrated robust anti-tumor activity in CD73-positive xenograft and syngeneic models, both as a monotherapy and in combination with anti-PD-1 therapy, while maintaining good tolerability and plasma stability.
