JACI-35.054 is an investigational conjugate vaccine developed for the treatment of Alzheimer's disease, specifically targeting tau pathology. It is designed to stimulate the immune system to generate antibodies against phosphorylated tau (pTau), a protein implicated in neurodegenerative processes characteristic of Alzheimer’s disease and related tauopathies. The vaccine uses a synthetic peptide sequence from pathological tau, linked to a carrier protein and formulated with two adjuvants, aiming to induce an immune response that targets both pathological and non-pathological forms of tau protein[2][6][8]. In clinical trials, JACI-35.054 demonstrated safety and induced antibody responses in early-stage Alzheimer’s patients; however, its antibody specificity for pathological forms of tau was less pronounced compared to ACI-35.030—a related liposomal vaccine using the same antigenic peptide but different delivery technology[2][6][8]. As a result, further development has prioritized ACI-35.030 over JACI-35.054[7].
