Jagged peptide is a synthetic peptide derived from the Notch ligand Jagged (typically Jagged1), specifically mimicking the Delta/Serrate/LAG-2 (DSL) domain required for Notch receptor activation. It functions as a Notch signaling agonist. In oncological research, particularly in glioblastoma multiforme (GBM), Jagged peptide is utilized to investigate the role of the perivascular niche in maintaining cancer stem-like cells (CSLCs). By activating Notch signaling, the peptide promotes the self-renewal, expression of stemness markers like CD133, and clonogenicity of GBM cells. While primarily employed as a research tool to study juxtacrine signaling and lateral inhibition, Notch-activating peptides are also explored for potential applications in regenerative medicine and hematopoiesis.
