Overview

JAM-1 nanobody (mE6) is a single-domain antibody (nanobody) developed by researchers at the Clem Jones Centre for Ageing Dementia Research at the Queensland Brain Institute, University of Queensland. It is designed to target junctional adhesion molecule 1 (JAM-1, also known as JAM-A or F11R), a transmembrane protein located at the tight junctions of epithelial and endothelial cells that regulates blood-brain barrier (BBB) integrity. mE6 binds to the membrane-proximal non-dimeric Ig-like domain (D2) near the linker region of JAM-1, creating steric hindrance that interferes with new dimer formation rather than directly disrupting existing cis-dimers. By weakening tight junctions, mE6 transiently increases paracellular permeability, serving as a BBB-weakening agent to enhance the delivery of therapeutic agents to the brain for the treatment of neurological disorders such as Alzheimer's disease.
