JANX007 is an investigational, protease-activated, masked T cell engager (TCE) designed to target prostate-specific membrane antigen (PSMA), a protein highly expressed in prostate cancer tumors and tumor vasculature. It is being developed for the treatment of metastatic castration-resistant prostate cancer (mCRPC). The molecule is a humanized trispecific protein containing PSMA-binding and CD3-binding domains, as well as an albumin-binding domain to extend half-life. A peptide mask inhibits CD3 engagement until it is cleaved by tumor-associated proteases in the tumor microenvironment, enabling localized T cell activation against PSMA-expressing cells while minimizing off-tumor toxicity. Preclinical and early clinical data show potent anti-tumor activity with improved safety compared to traditional TCEs[1][2][3][4][6].
