JANX008

JANX008 is a novel, double-masked bispecific T cell engager (TRACTr) designed to target the epidermal growth factor receptor (EGFR) on tumor cells and CD3 on T cells. The molecule incorporates masking domains for both the EGFR-binding and CD3-binding regions, which are cleaved by tumor-associated proteases in the tumor microenvironment. This design aims to minimize cytokine release syndrome (CRS) and healthy tissue toxicity while enabling potent, localized anti-tumor immune responses. Developed by Janux Therapeutics, JANX008 is being evaluated in Phase 1 clinical trials for advanced or metastatic solid tumors with high EGFR expression—including colorectal cancer, squamous cell carcinoma of the head and neck, non-small cell lung cancer, renal cell carcinoma, small cell lung cancer, pancreatic ductal adenocarcinoma, and triple-negative breast cancer[1][4][5][8]. Early clinical data show encouraging safety and efficacy signals with minimal severe adverse events[7][8].
