JANX009 is a bifunctional *Tumor Activated Immunomodulator (TRACIr)* developed for oncologic indications. It is designed to target both the **programmed death-ligand 1 (PD-L1) receptor** and the *costimulatory CD28 receptor* on T cells. The molecule is intended to selectively block PD-L1-mediated immunosuppression in the tumor microenvironment while simultaneously delivering CD28-mediated costimulatory signals—amplifying T cell-mediated anti-tumor activity within tumors but sparing healthy tissue. Its “double-masked” structure is engineered to restrict activation to the tumor site, aiming to improve safety and tolerability over earlier costimulatory approaches. JANX009 is being investigated primarily for solid tumors, with preclinical data supporting advancement into clinical studies. PD-L1 blocking activity is comparable to known PD-L1 inhibitors, and preclinical studies indicated that the molecule is well tolerated at high doses in non-human primates with extended half-life pharmacokinetics[3][5][8][6].
