Overview

Jaspisamide A is a marine macrolide toxin belonging to the trisoxazole family. It exhibits promising pharmacological properties due to its high-affinity and specific targeting of actin. Structurally, its trisoxazole ring interacts with actin subdomain 1, while its aliphatic side chain inserts into the hydrophobic cavity between actin subdomains 1 and 3. This binding site is analogous to that of the actin-capping domain of the gelsolin superfamily of proteins, suggesting that Jaspisamide A functions by severing and capping actin filaments, similar to gelsolin.
