Overview

Jasplakinolide is a naturally occurring cyclic depsipeptide originally isolated from the marine sponge *Jaspis johnstoni*. It is a potent inducer of actin polymerization and a stabilizer of F-actin filaments. By binding to F-actin with high affinity, it competes with phalloidin and promotes the nucleation of actin monomers into filaments, effectively lowering the critical concentration for polymerization. In therapeutic research, it is being explored for its ability to stabilize the endothelial cytoskeleton and enhance the microvascular barrier, particularly in the context of Acute Lung Injury (ALI). It also exhibits antifungal and anti-tumor properties, though its high toxicity has historically limited its clinical use. Recent investigations have focused on liposome-mediated delivery to target lung microvessels specifically.
