Overview

JB-128 is a synthetic small molecule analog of the natural product Salinopostin A (Sal A) [7911]. It is being developed as an antimalarial agent targeting the asexual blood stages of *Plasmodium falciparum*, with the schizont stage being particularly sensitive [7911]. JB-128 acts by inhibiting parasite α/β serine hydrolases, including a homolog of monoacylglycerol lipase (MAGL). Resistance to JB-128 is mediated by mutations in the mitochondrial PRELI domain-containing protein (PfPRELI), which is essential for parasite growth [7911]. JB-128 was co-developed through a collaboration involving Stanford University, Columbia University, and Calibr at Scripps Research Institute [7911].
