Overview

JBH492 is an investigational antibody-drug conjugate (ADC) developed by Novartis. It consists of a humanized IgG1 monoclonal antibody targeting the CC chemokine receptor 7 (CCR7), which is overexpressed in certain hematologic malignancies. The antibody is conjugated via a cleavable linker to the cytotoxic maytansinoid DM4 (ravtansine). Upon binding to CCR7 on cancer cells, JBH492 is internalized and trafficked to lysosomes where the DM4 payload is released, inhibiting tubulin polymerization and inducing cell death through mitotic arrest. The drug was primarily developed for the treatment of chronic lymphocytic leukemia and non-Hodgkin's lymphoma[1][2][3][4][8].
